Support for comparing populations with different pool-size? #4

Hello,

It looks like there is no support in popoolation2 for comparing data with
different pool-sizes. Do you plan on including any support for this, or do you
simply not recommend using popoolation2 unless one has similar sample sizes for
the two populations?

Thanks in advance!

FST sliding offers the option for entering different pool-sizes. This however
only has a minor influence on the results. You could test the influence of
different pool-sizes to the parameters.
Generally I would recommend to use high pool-sizes (>100). In this case I would
only expect a minor influence of different pool-size to the results.

Hi,

In addition to this, from the manual: "Most importantly the amount of DNA per
individual in a single pool should be constant."

Does this mean the number of reads per individual needs to be exactly the same?
i.e. must my samples be barcoded to ensure this or will this work with Illumina
reads from pooled DNA (the amount of DNA should be roughly similar in the
samples we have - but does it need to be exactly constant)?

Thanks,
Jon.
