Request for the following terms, as discussed on the OBI call. More terms will be added to the spreadsheet as they are reviewed. The spreadsheet follows the same ROBOT template as the assay harmonization project. https://docs.google.com/spreadsheets/d/1dkjrgPmjtu1svAm5FcHqUC8qqDlFzMwKbhtheBhSJM8/edit?usp=sharing

systematic evolution of ligands by exponential enrichment assay Parent: binding assay Definition: A binding assay that identifies protein-binding sites on nucleic acids by selecting high-affinity target ligands from a randomized pool. The process is repeated in rounds, then the bound nucleic acids are separated from the unbound nucleic acids and amplified by PCR.

mammalian 2-hybrid assay Parent: binding assay Definition: A binding assay that increases the expression of a reporter gene in mammalian cells by proteins of interest attached to two portions of the transcriptional activator, bringing those portions closer together.

protein localization assay Parent: assay Definition: An assay that determines the specific location of a protein. Subcellular localization is distinguished from tissue-based localization based on the type of microscopy applied.

tissue-based protein localization assay Parent: protein localization assay Definition: TBD

subcellular protein localization assay Parent: protein localization assay Definition: A protein localization assay that determines the specific subcellular location of a protein. The location is visualized through electron microscopy.

subcellular protein immunohistochemistry assay Parent: immunohistochemistry Definition: An immunohistochemistry assay that uses antibodies to display the specific subcellular location of proteins. Call comments: Was originally 'immunolocalization assay', changed label to be more specific.

DNAse footprinting assay Parent: TBD (binding assay?) Defintion: A ____ assay that determines protein-binding sites on DNA by identifying bound fragments that are protected from DNAse I-catalyzed hydrolysis.
