Question: myeloid dendritic cell lineage #2063

I am going to add a new term 'monocyte-derived cell' in CL and I realised that monocyte-derived dendritic cell CL:0011031 is a conventional dendritic cell. This is because it is a myeloid dendritic cell and myeloid dendritic cells in CL are a subclass of conventional dendritic cell: image Literature suggest that conventional DCs are myeloid dendritic cells PMID:30719026, and not the opposite, and therefore monocyte-derived dendritic cell would not be classified as cDCs. Is there a reason for CL to have it represented the opposite way?

Hi Aleix,

Sorry, I am mostly off this week, so can't respond at length right now. The relationship between conventional dendritic cells and myeloid DC is correct in CL according to a number of publications and has been discussed by me here and elsewhere.

The DC hierarchy in CL does need updating in a variety of ways and I would like to contribute to that.

What is prompting you to create a 'monocyte-derived cell' class, BTW?

@addiehl Hi Alex, our kidney experts requested 'monocyte derived cell" with description "A cell that is derived from monocytes and may differentiate into one of the myeloid cells including macrophages and dendritic cells." It looked like from CL that from such a superclass you would then have either monocyte derived dendritic cell (MoDC) or monocyte-derived macrophage. The recent Nature paper: https://www.nature.com/articles/s41577-020-00413-x was my source. Thank you for providing the input. From all the reading I had done, CL did not look correct in this case to my eyes, but I don't have your depth of knowledge in immunology.

Hi Alex, thank you for your answer. Additionally, some articles differentiate monocytes from monocyte derived cells (PMID:18160272):

About the DC hierarchy, in Figure 2, monocyte derived DCs are not classified as conventional DCs. I need to read more about this, but from what I have already read and your previous comments, they suggest that not all myeloid DCs are conventional DCs, and not all cDCs are myeloid DCs.
