In order to test whether the Gcn2 kinase induces autophagy through phosphorylation of eIF2a, we constructed a strain that expresses eIF2a with its phosphorylation site Ser52 substituted by ala- nine (eIF2a-S52A).

Phosphorylated eIF2a inhibits translation in a manner dependent on Gcn3, a component of eukaryotic translation initiation factor 2B (eIF2B) (Dever et al., 1993; Yang and Hinnebusch, 1996), followed by the up-regulation of the transcription factor Fil1 (Duncan et al., 2018).
