Replacement of all fission yeast CTD Ser7 residues with alanine results in derepression of pho1, pho84, and tgp1 expression in phosphate-replete cells, whereas replacement of all Thr4 residues with alanine causes pho1 and pho84 hyperrepression (12).

We hadshown that an active site mutation in the N-terminal kinase domain that abolishes IP8 synthesis results in hyperrepression of pho1 expression, while an active site mutation in the pyrophosphatase domain (that elevates IP8 levels) causes pho1 derepression in a manner that requires CPF subunits and Rhn1 (25).

Conversely, the PHO genes are hyperrepressed in cells bearing loss-of-function mutations in components of the 3′-processing/termination machinery (12, 13).
