I am trying to demultiplex a minion run which I used 6 different barcodes (kit SQK-RBK004)
For such a reason I can't understand it recognise perfectly the first 5 barcodes but fails to recognise the 6. I looked into the different files of separated barcodes using grep and I can find barcode 6 mainly in none but also here and there in all the other 5 subsets.
Do you have any idea of why this is happening? And how to fix it?
I used porechop on a smaller subset of samples and it worked perfectly, is size a problem?
