Problem in accessing API/Proxy.svc/ajax/js file at runtime

Hi Team,

I am getting problem in below js file.
<script src="../API/Proxy.svc/ajax/js" type="text/javascript"></script>

Here above file is not loading in my page. So please provide me solution.

This is done by scriptmanager control automatically. See the code in: 
ScriptManagerControl.ascx
