Cutoff for Scissor-selected cell #67

Hello,

I have a question regarding the cutoff value of 0.2 for the maximum number of Scissor-selected cells. This means that the number of Scissor-selected cells should not exceed 20% of the total cells in the single-cell dataset. What is the rationale behind setting this value? Biologically, this implies that the cell type being tested will contribute equally to the disease (~20% of the cell population). However, in reality, a large number of T cells are likely to contribute more to immune-related diseases compared to a smaller number of neuronal cells. I would appreciate your insights on this matter. Thanks!

Hi Siqi

Your consideration is correct. This cutoff is not a fixed parameter.

We used cutoff = 0.2 to show the effectiveness of Scissor. In real applications, you can change this number according to your requirement.
