We're sharing a keepass file on a webserver with authenicated access. Would this lookup plugin support such a setup? Would this work in AWX/AAP/Tower?

To use this collection, you need to have a Keepass file on Ansible machine. I have never used Tower, but I think if the collection works in terminal, then it will work in Tower (try it).
