Cmax different from literature #1

Hi Wang Wei, I saw your simulation on carvedilol for (Manuel Ibarra, Cristian Valiante, Patricia Sopeña, et al. Integration of in vitro biorelevant dissolution and in silico PBPK model of carvedilol to predict bioequivalence of oral drug products. European Journal of Pharmaceutical Sciences, 2018,118: 176-182).

I read your thread on Github and downloaded your file to try on the simulation. In addition, I also created my own file. However, one problem with my simulation is that the Cmax is always high and the range 2.5 - 97.5% is very large.

I tried repeating but I could not find out what went wrong. I want to ask if there are any flaws in my simulation? https://github.com/yixintan1009/PK-Sim-File.git

Hi, Yixin Tan,

I have checked your file and found some problems.

First, in the Administration Protocols , your Administration type is User Defined. This should be changed to Oral if you want to simulate an oral dosage. Even if you define a Target organ as stomach or intestine under User Defined, this is still wrong because the organs mean the orangs themselves, not the lumen of the organs.

Second, the setting in your population is improper. The Demographics show the age range is between 0 to 100, which results in a large body weight distribution between less than 10 to 120 kg and eventually enlarge the drug concentration variation.
